    SEE ALSO...

CSP Antibody (H-3) is a mouse monoclonal IgG1 kappa light chain antibody that detects CSP protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-CSP antibody (H-3) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Cysteine string proteins (CSPs) play a crucial role in regulating synaptic transmission, as CSPs are associated with synaptic and secretory vesicles and are involved in calcium-regulated exocytosis. CSP contains a J-domain that interacts with heat shock protein 70 (HSP70) and heat shock cognate protein 70 (HSC70) chaperones, facilitating proper folding of proteins essential for vesicular exocytosis. Additionally, CSPs are characterized by a palmitoylated cysteine-rich string region that aids in membrane targeting, which is vital for their function in synaptic vesicle dynamics. CSP interaction with receptor-coupled trimeric GTP-binding proteins and N-type calcium channels underscores CSP importance in fine-tuning neurotransmission. Notably, two isoforms of CSP have been identified: CSP1, which is predominantly found in granular fractions of insulinoma cells, and CSP2, a truncated variant that is more prevalent in membrane and cytosolic fractions. Differential expression of these isoforms in various mammalian cell lines, including rat brain, highlights CSP′s role in cellular signaling and neurotransmitter release.

Why is the size of western blotting band 34 kDa ? In database, CSP/Dnajc5 protein size is 22 kDa. is this band really CSP? How can you sure, even the size is completely different ?

    Asked by: JKkim
Thank you for your question. We have observed CSP in different tissue extracts at approximately 35 kDa using this antibody, CSP (H-3): sc-137128, as well as another antibody to the same target with a different epitope, CSP (16): sc-136468, (https://www.scbt.com/scbt/product/csp-antibody-16). This observed molecular weight is also consistent with a reference for CSP reported in the literature, on NCBI PubMed, PMID: 7666205.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with CSP (H-3): sc-137128?

    Asked by: DefinitelyNotMatt
We recommend using one of our our exclusive Mouse IgG Binding Proteins conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems. A complete list of available binding proteins is available on our website here: https://www.scbt.com/scbt/browse/support-products-mouse-igg-binding-proteins/_/N-ecrety
