SEE ALSO...

Cripto Antibody (H-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects Cripto protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-Cripto antibody (H-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Cripto protein, also known as cripto-1, is encoded by the teratocarcinoma-derived growth factor (TDGF)-1 gene and plays a crucial role in early embryonic development, particularly during gastrulation when Cripto is first expressed in the forming mesoderm. Cripto expression later becomes restricted to the truncus arteriosus of the developing heart, indicating Cripto′s involvement in the progression of epiblastic cells that contribute to mesoderm formation. In adult tissues, Cripto is found at low levels in the spleen, heart, lung, and brain, but Cripto overexpression is notably associated with human gastric and colorectal carcinomas, highlighting Cripto′s potential as a biomarker for cancer progression and a target for therapeutic intervention.

Can Cripto (H-10): sc-376448 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Asked by: Germaine
Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, Cripto (H-10): sc-376448 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 5 out of 5 by from Great results using Santa Cruz Cr-1 antibody. We have been investigating the expression of Cripto-1 in reproductive tissues and particularly in trophoblast cells. Our results showed gene and protein up-regulation of this factor in placentation associated with abnormal trophoblast invasion (creta placentas, Bandeira et al: BioMed Research International, 2014). Cr-1 silencing decreased the invasive capacity of the trophoblast cell lineages and suppressed specific signaling pathways, also found in cancer cells. The putative role of this factor in determining diagnosis and prognosis of conditions associated with abnormal invasion of the trophoblast is, therefore, promissory.
