COX4 Antibody (F-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects COX4 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). COX4 (F-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The cytochrome c oxidase (COX) complex, of which COX4 is a crucial component, serves as the terminal oxidase in the mitochondrial respiratory chain, facilitating the transfer of electrons from cytochrome c to molecular oxygen, thereby playing a vital role in cellular respiration and energy production. The structure of COX is particularly important as COX consists of a heteromeric assembly of both mitochondrial-encoded and nuclear-encoded subunits, which is essential for enzymatic function. COX4, being a nuclear-encoded subunit, is integral in modulating the activity of the COX complex, ensuring efficient energy production in cells. COX4 is ubiquitously expressed in adult human tissues, with highest expression levels found in the pancreas, and moderate levels in the heart, skeletal muscle, and placenta, highlighting COX4′s essential role in various physiological processes.

Thank you for your question. COX4 Antibody (F-8) is only recommended for detection of COX4 of human origin by WB, IP, IF, IHC(P) and ELISA. Based on our tests we do not recommend it for murine cells or tissues.

Can you recommend a positive control to use with COX4 (F-8): sc-376731 monoclonal antibody for Western blot?

We recommend A-431 (sc-2201), MOLT-4 (sc-2201), Raji (sc-364236), MCF7 (sc-2206) and A-673 (sc-2414) whole cell lysates to use as positive control.

Rated 1 out of 5 by from No band!I used fresh antibody dilution (1:100 and 1:50) on 10ug HUVEC cell lysate and I did not get any signal in western blotting. Conversely, I detected a sharp band with anti-HADHA (sc-374497) on the same blot.

Rated 1 out of 5 by from too many nonspecific bandsI have been using COX4 antibody from Santa Cruz for mitochondrial fractionation (Western blot). Since the older product was no longer available, I ordered this version of antibody. And there are just too many of the nonspecific bands all over, including 30 kD band signal, which is virtually the strongest signal in my membrane. I wonder how others use this antibody.
