COP1 (B-12) is a mouse monoclonal IgG1 kappa light chain antibody that detects COP1 protein of mouse, rat and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP) and enzyme-linked immunosorbent assay (ELISA). COP1 (B-12) antibody is available as both non-conjugated form and multiple conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC) and multiple Alexa Fluor® conjugates. COP1 (Constitutive photomorphogenesis protein 1), also designated RFWD2 (ring finger and WD repeat domain 2) or RNF200 (ring finger protein 200), is an E3 ubiquitin ligase protein that mediates ubiquitination and degradation of target proteins such as c-Jun and p53. COP1 functions as a component of the DCX DET1-COP1 ubiquitin ligase complex which consists of RBX1, DET1, DDB1, CUL4A and COP1. COP1 localizes to the cytoplasm and nucleus, with primary expression in testis, placenta, heart and skeletal muscle. COP1 acts as a potent inhibitor of p53-dependent transcription and apoptosis but, when phosphorylated by Atm (ataxia telangiectasia mutated) in response to DNA damage, the COP1-p53 complex is disrupted and p53 is allowed to exert its pro-apoptotic properties. COP1 shows overexpression in ovarian and breast cancers, suggesting a role in tumorigenesis.

When should the TransCruz reagent sc-166799 X be used instead of the standard form sc-166799?

Asked by: Randy McDonald
Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
