Connexin 43 Antibody (D-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects connexin 43 in mouse, rat, human, and canine samples through western blotting (WB) and immunoprecipitation (IP). Anti-connexin 43 antibody (D-7) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Connexin 43, also known as GJA1, is a critical gap junction protein that forms hexameric structures called connexons, which cluster to create gap junctions facilitating intercellular communication. This communication is vital for maintaining tissue homeostasis and coordinating cellular responses, particularly in cardiac and neuronal tissues. Notably, connexin 43 is subject to various post-translational modifications, including phosphorylation, which can significantly influence function. During mitosis, connexin 43 undergoes specific phosphorylation that correlates with a temporary loss of gap junction intercellular communication, highlighting dynamic roles in cellular signaling and regulation of cell proliferation. In pathological conditions, such as cancer, altered expression and function of connexin 43 can lead to impaired gap junction communication, contributing to tumor progression and metastasis. Anti-connexin 43 antibody (D-7) effectively detects this crucial protein, making connexin 43 monoclonal antibody (D-7) an essential tool for researchers studying cell communication, cancer biology, and physiological roles of gap junctions.

I would like to use connexin 43 Antibody (D-7): sc-13558 for double staining with another mouse monoclonal primary antibody. Is this possible?

Thank you for your question. In cases like this, we recommend using our directly conjugated mouse monoclonal primary antibodies. Currently, sc-13558 is not available directly conjugated, but we have an alternative connexin 43 antibody (sc-271837) that is available conjugated to HRP, FITC, PE, Alexa Fluor® 488 and Alexa Fluor® 647. Rated 5 out of 5 by from Worked for WB on human cellsThis was my first time blotting for connexin 43. I had a faint but clear band from human breast cancer cell lysates. I'm not sure if the expression level is just low in these cells, but it may just be that you need a higher concentration of antibody and fresh lysates.

Rated 2 out of 5 by from Not so good for IF in tissue cryosections.I received a sample of this and tested it (1:100 dilution) with rat tissue cryosections fixed in ice-cold methanol; it did not work. The conjugated Ab (-conjugated with fluorophore such as 488) would probably be better. The F-7 antibody works well in IF.
