Clusterin-α Antibody (B-5) is a mouse monoclonal IgG1 kappa light chain antibody engineered to target Clusterin-α of human origin, specifically recognizing an epitope mapped between amino acids 432-449 at the C-terminus. Clusterin-α monoclonal antibody (B-5) is validated for use in Western Blot (WB), Immunoprecipitation (IP), Immunofluorescence (IF), Immunohistochemistry on paraffin-embedded tissues (IHCP), and ELISA. Clusterin-α, also known as apolipoprotein J (ApoJ), is a multifunctional secretory glycoprotein involved in various critical physiological processes including immune regulation, cell adhesion, lipid transportation, tissue remodeling, membrane recycling, and cell-cell interactions. The role of Clusterin-α as a complement lysis inhibitor is essential for protecting cells from complement-mediated damage, thereby maintaining immune homeostasis. Additionally, Clusterin-α interacts with numerous proteins to influence processes such as apoptosis, neuroprotection, and the aggregation of amyloid-beta peptides, which are implicated in the pathogenesis of Alzheimer′s disease. The heterodimeric structure of Clusterin-α, consisting of α and β subunits linked by disulfide bonds, is crucial for stability and functional versatility. Post-translational modifications, including glycosylation and proteolytic cleavage, are vital for proper folding, secretion, and activity of Clusterin-α, affecting interactions and functional outcomes in different cellular contexts. Overexpression of Clusterin-α has been observed in various pathological conditions, including cancer, where Clusterin-α is associated with tumor progression and metastasis, as well as in neurodegenerative diseases, where Clusterin-α may play either protective or pathological roles depending on the context. By specifically targeting the C-terminal region of Clusterin-α, Clusterin-α (B-5) monoclonal antibody facilitates detailed studies of expression, localization, and interactions within different biological systems, providing valuable insights into roles in health and disease. Clusterin-α mouse monoclonal antibody (B-5) is recommended for investigating immune regulation, cancer biology, neurodegeneration, and the development of therapeutic strategies.

Thank you for your question. Clusterin-alpha Antibody (B-5) is only recommended for detection of Clusterin-α of human origin by WB, IP, IF, IHC(P) and ELISA. It is not suitable for cell or tissue samples from mouse.

The protein band detected as clusterin-alpha percursor corresponds to the percursor containing both the N-signal and alpha and beta sequences right?

Thank you for your question. Clusterin-α Antibody (B-5): sc-5289 recognizes amino acids 432-449, which correspond to Clusterin alpha chain. As such, the antibody detects any form of Clusterin containing this sequence.

I want to ask why many clusterin antibody are clusterin-alpha chain antibody? not alpha and beta chain together, or only beta chain? Does it really can identify clusterin protein? Don't know how to choose the suitable clusterin antibody.

Clusterin is synthesized as a 449 amino acid polypeptide that is post-translationally cleaved at an internal bond between Arg 227 and Ser 228. Two subunits, α and β, are associated through disulfide bonds. The β subunit (also called ApoJα) corresponds to residues 23-227. The α subunit (also called ApoJβ) corresponds to residues 228-449. Based on epitope mapping of the antibody, it is possible to make antibodies that are specific to either the alpha or beta chain. If the epitope has not been mapped, then the antibodies may be recommended for detection of Clusterin protein.

hello, i want to detect to nuclear clusterin as IHC-P and WB. So can i use Clusterin-α Antibody (B-5): sc-5289 ? Because i see H-330 doesent product anymore.

How can Clusterin-α (B-5): sc-5289 mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. Clusterin-α (B-5): sc-5289 monoclonal antibody is currently available conjugated to phycoerythrin (sc-5289 PE), fluorescein (sc-5289 FITC), Alexa Fluor® 488 (sc-5289 AF488) or Alexa Fluor® 647 (sc-5289 AF647)
