claudin-1 Antibody (A-9) is a mouse monoclonal IgG2b kappa light chain antibody targeting an epitope between amino acids 168-207 at the C-terminus of human claudin-1, with reactivity across human, mouse, rat, equine, canine, and bovine species by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Claudin-1, also known as CLDN1, is an essential member of the claudin family, which forms the backbone of tight junctions in epithelial and endothelial cells, regulating paracellular transport. Structurally, claudin-1 spans the membrane with four transmembrane domains and two extracellular loops, which are critical in forming tight junction strands that create selective barriers between cells. This unique structure allows claudin-1 to participate in maintaining cellular polarity and controlling ion permeability, vital functions in organs like the liver and kidney, where claudin-1 is most abundantly expressed. The structural configuration of claudin-1 also facilitates interactions with other tight junction proteins, such as occludins and junctional adhesion molecules, enabling complex tissue compartmentalization. Mutations in claudin-1 are linked to neonatal ichthyosis-sclerosing cholangitis syndrome, highlighting its role in tissue integrity. Anti-claudin-1 antibody (A-9) is available in both non-conjugated and multiple conjugated forms, making it versatile for research across various species and applications.

can SC-166338 be biotinylated? I have an EZ-Link NHS-PEG 4-Biotin Biotinylation kit cat#21329

We are not familiar with that exact conjugation kit, however the standard formulation of this antibody does contain gelatin as a stabilizing protein, and the gelatin may interfere with the conjugation process for the antibody. We can offer this antibody without gelatin, or conjugated to Biotin, as a special order. Please contact our Technical Service Department for more information or to request a quote. Can claudin-1 Antibody (A-9): sc-166338 be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Yes, claudin-1 Antibody (A-9): sc-166338 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat.
