Clathrin LC Antibody (CON.1) is a mouse monoclonal IgG2b kappa light chain antibody that detects Clathrin LC protein across multiple species through western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Anti-Clathrin LC antibody (CON.1) comes in non-conjugated form and various conjugates, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® options. Clathrin plays a crucial role in intracellular transport, particularly in receptor-mediated endocytosis, facilitating receptor protein engulfment into clathrin-coated vesicles. Clathrin LC protein resides primarily in the cytosol, assembling into a triskelion structure composed of three heavy chains and three light chains. This structural arrangement enables coated vesicle formation for protein and lipid transport within cells. Light chains, specifically Clathrin light chain A (LCA) and Clathrin light chain B (LCB), regulate triskelion self-assembly onto membranes, influencing vesicle formation efficiency. LCA and LCB, encoded by distinct genes sharing 60% homology, demonstrate similar functional characteristics, including alternative mRNA splicing for tissue-specific isoforms. Brain-specific light chain isoforms contain inserted sequences forming higher molecular weight variants, essential for neuronal function and signaling.

What is the recommended fixation method for immunofluorescence staining with Clathrin LC (CON.1): sc-12735 monoclonal antibody?

Asked by: Germaine
Thank you for your inquiry. We recommend fixing cells for 5 minutes in -10°C methanol and allowing to air dry. The full protocol can be viewed here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining
