cGAS Antibody (D-9) is a mouse monoclonal IgG2a kappa light chain antibody that detects cGAS of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-cGAS antibody (D-9) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. cyclic GMP-AMP synthase (cGAS) plays a crucial role in innate immune response by acting as a sensor for cytosolic DNA, which is often indicative of viral infection. Upon binding to foreign DNA, cyclic GMP-AMP synthase catalyzes the synthesis of cyclic GMP-AMP (cGAMP) from ATP and GTP, a second messenger that activates the stimulator of interferon genes (STING) pathway, leading to the production of type I interferons and other pro-inflammatory cytokines. This process is vital for initiating an effective antiviral response, highlighting the importance of cyclic GMP-AMP synthase in immune surveillance and defense against pathogens. cyclic GMP-AMP synthase is expressed in various cell types, including the monocytic cell line THP1, and exists as two alternatively spliced isoforms, with its gene located on human chromosome 6q13.

Thank you for your question . Yes we also recommend the antibody for IF,IHC and FCM. The antibody has only been validated for human. There is a 64% homology and 56% homology in rat .

Is this can be used to detect mouse sample?

Thank you for your question. As indicated in the product description, cGAS Antibody (D-9) is only recommended for detection of cGAS of human origin by WB, IP, IF, IHC(P) and ELISA. It is not recommended for detection of cGAS of mouse and rat origin.

I am using cGAS (D-9): sc-515777 for immunofluorescence, what dilution of this antibody should I use?

Rated 5 out of 5 by from This Ab can recognize mouse cGASI have shown by shRNA knockdown that this antibody (D-9) recognizes mouse cGAS. A band of the same size was strongly detected in human cells (BJ-5ta). Unfortunately, the C-1 antibody did not detect a band of the correct size.

Rated 1 out of 5 by from Not recognizing mouse cGAS!I used cell lysates from primary bone-marrow derived macrophages from C57BL/C6 WT and cGAS KO (ordered from Jackson laboratory cat# 026554) mice for western blot and got a band around 60 kDa in both samples. Clearly, this antibody is not detecting cGAS!

Rated 1 out of 5 by from Antibody is not detecting cGAS!The antibody is detecting a protein a correct mass of about 60 kDa. However, the same band is detectable in three different human cGAS-/- clones (BLaER1 cells). The antibody is not specific!

Rated 4 out of 5 by from Works goodI tried a free trial sample of this antibody. I validated this antibody by comparing HeLa with HEK293. HEK293 does not express cGAS so it can be used as a 'KO validation'. Used as 1:1000 in 5% milk TBS-T.
