CENPB encodes centromere protein B (CENP-B), a sequence-specific DNA-binding factor that recognizes the CENP-B box within α-satellite repeats and helps organize centromeric chromatin. CENP-B contributes to kinetochore assembly and chromosome segregation by supporting centromere architecture and higher-order heterochromatin organization during mitosis. Through its roles in centromere maintenance and cell-cycle progression, altered CENPB function or centromere dysregulation is relevant to genome instability phenotypes, including aneuploidy and chromosomal aberrations observed across diverse cancers. CENP-B is also a well-established autoantigen in anti-centromere antibody responses, linking centromere biology to autoimmune-associated cellular phenotypes.
CENP-B CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the CENPB gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the CENPB together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the CENPB open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish CENP-B protein expression.
This CRISPR knockout system enables efficient generation of CENPB-deficient cell models for investigation of CENP-B signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
