Cyclin-dependent kinase inhibitor 3 (CDKN3), also known as Cdk2-associated dual specificity phosphatase, cyclin-dependent kinase interactor 1 (CDI1), CIP2, KAP, or KAP1, is a member of the protein-tyrosine phosphatase family and plays a crucial role in cell cycle regulation. CDKN3 is primarily located in the nucleus, where CDKN3 is essential for controlling the progression of the cell cycle by regulating the activity of cyclin-dependent kinases (CDKs). Nuclear localization of CDKN3 allows for precise modulation of CDK activity, which is vital for maintaining proper cell division and preventing uncontrolled proliferation. CDKN3 interacts with and dephosphorylates Cdk2 kinase, thereby inhibiting activation and ensuring that cells do not prematurely enter the S phase of the cell cycle. Dysregulation of CDKN3 has been implicated in various cancers, as mutations, overexpression, or deletions of the CDKN3 gene can lead to aberrant cell cycle progression and tumorigenesis.
