CDKL5 Antibody (D-12) is a mouse monoclonal IgG2b kappa light chain antibody that detects CDKL5 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-CDKL5 antibody (D-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CDKL5, which consists of 1,030 amino acids, is a member of the CMGC Ser/Thr protein kinase family and is primarily expressed in the brain, lung, kidney, prostate, ovary, placenta, pancreas, and testis. CDKL5 plays a crucial role in cell cycle regulation by interacting with cyclins and cyclin-dependent kinases to phosphorylate key substrates necessary for cell cycle progression. CDKL5 is implicated in several neurological disorders, including X-linked infantile spasm syndrome and Rett syndrome, highlighting CDKL5′s significance in both normal cellular function and disease pathology. CDKL5 (D-12) antibody′s ability to effectively detect CDKL5 across multiple species and applications makes this reagent an invaluable tool for researchers studying cell cycle dynamics and related disorders.

I am using this antibody for WB for WT and KO mice however it is showing bands in both lanes (for WT and KO) at the expected MW of the protein. Has this problem occured before? Behavioural tests on these mice do show that they are KO.

HELLO, I am using this antibody for WB, but i am getting lots of unspecific binding, the dilution I am doing is 1:2500, and for the blocking I am using milk. any suggestions or recommendations to get better results?

Thank you for your question. For western blotting experiments with this antibody we recommend a starting dilution of 1:100, and a dilution range between 1:100-1:1000. If the issue persists with dilutions within the recommended range please contact our technical service team for in-depth troubleshooting of your experiment.

Has this antibody been tested in IF on WT and Knock Out cells or in cells where CDKL5 has been knocked down? Because in my hands, it seems to give good nuclear signal even in cells where CDKL5 has been knocked out using CRISPR or knocked down using siRNA.
