SEE ALSO...

Cdk9 Antibody (F-6) is a mouse monoclonal IgG1 kappa light chain antibody designed to target Cyclin-dependent kinase 9 (Cdk9), also known as PITALRE or CDC2L4. anti-Cdk9 antibody (F-6) specifically recognizes an epitope mapping between amino acids 343-372 at the C-terminus of the human Cdk9 protein, ensuring precise and reliable detection. Cdk9 is a pivotal member of the cyclin-dependent kinase family, characterized by its conserved kinase domain that facilitates ATP binding and substrate phosphorylation, which are essential for enzymatic activity. The structural integrity of Cdk9, particularly within its C-terminal region, is crucial for interaction with cyclin partners such as Cyclin T1, forming the positive transcription elongation factor b (P-TEFb) complex. This complex plays a vital role in regulating transcription elongation by phosphorylating the C-terminal domain of RNA polymerase II, thereby enabling the expression of genes involved in critical cellular processes like cell cycle progression, differentiation, and apoptosis. The structural domains of Cdk9 are integral to function, as they determine substrate specificity and regulatory mechanisms, making accurate detection of conformation and interactions essential for understanding roles in cellular homeostasis and disease pathology. Additionally, Cdk9 is involved in phosphorylating the retinoblastoma protein, linking to the control of cell cycle and cellular proliferation. Dysregulation of Cdk9 structure and function is associated with various diseases, including cancer and viral infections such as HIV, where Cdk9 is necessary for transcriptional activation mediated by HIV Tat protein. Cdk9 (F-6) antibody demonstrates broad species reactivity, effectively detecting Cdk9 from mouse, rat, and human origins through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA), and is also reactive with additional species including equine, canine, bovine, and porcine.

What application is the blocking peptide sc-376646 P appropriate for?

Asked by: cjMara
Thank you for your question. The blocking peptide is intended for use as a negative control, by pre-adsorbing the mouse monoclonal antibody against the antigen. For full protocol details, please contact our Technical Services Department or view our online protocol here: https://www.scbt.com/scbt/resources/protocols/peptide-neutralization
