CDC37 encodes Cdc37, a conserved HSP90 co-chaperone that scaffolds and stabilizes a broad set of protein kinases, supporting their maturation, conformational maintenance, and signaling competence. By coordinating HSP90–client interactions, Cdc37 influences key processes including cell-cycle progression, stress responses, and signal transduction across MAPK, PI3K–AKT, and other kinase-driven pathways. Perturbation of CDC37 can alter proteostasis of oncogenic and developmental kinases, reshaping phosphorylation networks and downstream transcriptional programs. As a regulator of kinase homeostasis, CDC37 is frequently studied in contexts of aberrant proliferation, proteotoxic stress, and pathway rewiring relevant to cancer biology and other kinase-associated diseases.
Cdc37 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the CDC37 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the CDC37 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the CDC37 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Cdc37 protein expression.
This CRISPR knockout system enables efficient generation of CDC37-deficient cell models for investigation of Cdc37 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
