CD68 Antibody (E-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects CD68 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), flow cytometry (FCM), and enzyme-linked immunosorbent assay (ELISA). Anti-CD68 antibody (E-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CD68 is a member of a family of acidic, highly glycosylated lysosomal glycoproteins (LGPs), which includes lamp-1 and lamp-2, and shares homology with mouse antigen macrosialin. CD68 plays a crucial role in the immune system, primarily functioning as a marker for macrophages and other myeloid cells, which are essential for phagocytosis and cellular debris clearance. CD68 is predominantly located in cytoplasmic granules and cytoplasm of various non-hematopoietic tissues, such as liver and kidney tubules and glomeruli, and appears on the surface of macrophages, monocytes, neutrophils, basophils, and large lymphocytes, albeit to a lesser extent. CD68 expression on these immune cells is vital for their function in immune response, aiding in pathogen recognition and uptake of apoptotic cells, thereby contributing to tissue homeostasis and inflammation resolution.

Does the antibody sc-17832 require antigen retrieval?

Thank you for your question. We recommend performing antigen retrieval via Heat treatment: Place slides in a container and cover with 10 mM sodium citrate buffer, pH 6.0 (sc-294091); or with 50 mM glycine-HCl buffer (sc-29096), pH 3.5, with 0.01% (w/v) EDTA (sc-29092). Heat at 95° C for 5 minutes. Top off with fresh buffer and heat at 95° C for 5 minutes (optimal incubation time may vary for each tissue type). Allow slides to cool in the buffer for approximately 20 minutes. Wash in deionized H2O three times for 2 minutes each. Aspirate excess liquid from slides.

Can CD68 (E-11): sc-17832 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, CD68 (E-11): sc-17832 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
