SEE ALSO...

CD40 Antibody (HM40-3) is a hamster monoclonal IgM antibody that detects CD40 protein of mouse and rat origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and flow cytometry (FCM). Anti-CD40 antibody (HM40-3) is available in a non-conjugated form, providing researchers with a versatile tool for studying the CD40 signaling pathway. CD40 plays a crucial role in the immune response, particularly in the activation and differentiation of B cells. CD40 is primarily located on the surface of B cells and interacts with CD40 ligand (CD40L) expressed on activated T cells, facilitating essential cell-to-cell communication. This interaction is vital for B cell activation, leading to clonal expansion and the production of antibodies. The importance of CD40 in immune regulation is underscored by its involvement in immunoglobulin class switching and the secretion of IgE, which are critical processes in the adaptive immune response. Furthermore, the CD40 signaling pathway is mediated by downstream effectors such as CRAF, which is part of a family of proteins that includes TRAF1 and TRAF2, known for their roles in regulating tumor necrosis factor receptor signaling and activating the NFκB transcription factors. Understanding the function and interactions of CD40 is essential for elucidating mechanisms of immune responses and developing therapeutic strategies for immune-related diseases.

This antibody is giving another band at around 54kD (other than the 43kD band). The results are consistent. Is its another isoform of CD40, that it is detecting? Its a virus-induced CD40 expression that we are checking.

Asked by: DEBANJANA
Thank you for your question. According to the literature, CD40 is a glycoprotein that could be detected at higher molecular weights.

To do the Western blot of this antibody, how can I get the HRP conjugated anti hamster IgM? Would do please tell me the order information if possible?

Asked by: Ying2626
CD40 Antibody (HM40-3) is a Armenian hamster monoclonal IgM. Unfortunately, we do not currently offer an isotype-specific secondary antibody that is anti-Armenian hamster IgM-HRP. You could try using our mouse anti-Armenian hamster IgG-HRP: sc-2789, which may also be suitable for detection of IgM.

what if the antibody is kept at room temperature for more than 2 days accidentally? will it still work?

Asked by: debby
Thank you for the question. We suggest storing this antibody at +4°C - 8°C in the fridge. If stored accidentally at room temp. for 2 days the product likely will not be harmed. We still suggest evaluating the antibody with a positive control and verify the obtained data.

How can CD40 (HM40-3): sc-20010 mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

Asked by: jerojero
In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is available conjugated to phycoerythrin (sc-20010 PE) or fluoroscein (sc-20010 FITC).
