CD34 Antibody (ICO115) is a mouse monoclonal IgG1 kappa light chain antibody that detects CD34 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). CD34 (ICO115) antibody is available in both non-conjugated and various conjugated forms, including agarose, phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CD34 is a heavily glycosylated transmembrane glycoprotein that plays a crucial role in identifying and isolating hematopoietic stem and progenitor cells, as CD34 is predominantly expressed on their surface. This expression is particularly significant because CD34 serves as a marker for the most primitive stem cells, which are essential for maintaining hematopoiesis and tissue regeneration. As these stem cells differentiate into lineage-committed progenitors, CD34 expression gradually diminishes, highlighting CD34′s importance in stem cell biology and potential therapeutic applications. CD34 is found on small-vessel endothelial cells and certain cells in fetal and adult nervous tissue, further underscoring CD34′s diverse roles in various biological contexts. The cytoplasmic domain of CD34 contains consensus sites for phosphorylation by activated protein kinase C and other kinases, which may influence signaling pathways critical for cell adhesion and migration.

Good morning, I would like to confirm whether the ICO115 clone of CD34 has been published for use in dogs, like the previous C-18 clone.

Thank you for your question. Based on our tests with the antibody, we only recommend CD34 (ICO115): sc-7324 for detection of CD34 of mouse, rat and human origin by WB, IP, IF and FCM. We do not have any internal or customer data on viability in canine cell or tissue samples.

I would like to know if this antibody (anti-CD34 sc-7324) was used or tested on dermal vascular tumors in canines. Thank you.

Thank you for your question. Anti-CD34 Antibody (ICO115) is only recommended for detection of CD34 of mouse, rat and human origin by WB, IP, IF and FCM. We do not have any data concerning the antibody's viability in canine cell or tissue samples.

Does this antibodies works with Bovine cells ? we need to isolate mesenchymal cells from bovine adipose tissue.

I am looking for a good antibody for hematopoietic cells in dogs, has been used for it?

Thank you for your question. We are not aware of any citations using this CD34 Antibody (ICO115): sc-7324 in samples of canine, or dog origin. Therefore, we do not know if it will react with CD34 in dog. We do offer an alternative antibody that is recommended for detection of CD34 of canine (dog) origin by WB and FCM. That is our mouse monoclonal, CD34 Antibody (1H6): sc-53511. A link to the product information is provided below. Someone has tested the antibody on live cell culture without fixation? I have embryoid body cultures to stain and I'm looking for the right antibody which would penetrate deep in the bodies and not require fixation/permeabilization. Thanks

Thank you for your question. We have not tested this CD34 Antibody (ICO115): sc-7324 on live cell culture without fixation or permeabilization. This antibody is very well cited in the primary literature, with many citations using the antibody for immunofluorescent staining or immunhistochemistry. We would recommend consulting with the primary literature to see if anyone else has used this clone for your same intended research application.

What negative control should be used when using the conjugated version of CD34 Antibody (ICO115): sc-7324 in flow cytometry?

We recommend using the isotype specific normal mouse IgG conjugated to the same fluorophore. For example, if you are using sc-7324 PE, then you would want to use normal mouse IgG1 PE, sc-2866.
