CD3 Antibody (17A2) is a rat monoclonal IgG2b antibody designed to recognize mouse CD3, recommended for assays like immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). CD3, also known as the T cell co-receptor complex or the TCR-CD3 complex, is located on the surface of T lymphocytes where CD3 plays a crucial role in T cell receptor (TCR) signaling. This localization is essential for the activation and regulation of T cells, enabling T cells to respond effectively to foreign antigens. The CD3 complex consists of five invariant polypeptide chains, gamma, epsilon, delta, zeta, and eta, that form three distinct dimers, each containing immunoreceptor tyrosine-based activation motifs (ITAMs). These ITAMs are vital for initiating intracellular signaling cascades by recruiting protein kinases such as ZAP-70 and Syk, which are necessary for T cell activation and proliferation. By targeting the CD3 complex, anti-CD3 antibody (17A2) allows researchers to study T cell activation, signal transduction, and immune response mechanisms. CD3 monoclonal antibody (17A2) binding to CD3 on mouse cells makes CD3 (17A2) antibody an indispensable tool for immunological research, enabling detailed analysis of T cell function and regulation in various experimental settings.

Can CD3(17A2) be used in FACS followed by T cell functional studies? Will it activate T cells or have no effect on T cells? Could all CD3 antibodies activate T cells? Thank you so much!!!

Can CD3 (17A2): sc-18843 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, CD3 (17A2): sc-18843 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
