CD223 Antibody (D-8) is a mouse monoclonal IgG2b kappa light chain antibody that detects CD223 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). CD223 (D-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. CD223, also known as Lymphocyte activation gene-3 (LAG-3), plays a crucial role in immune regulation by acting as a high-affinity ligand for major histocompatibility complex (MHC) class II molecules, which are essential for T cell activation and function. Located on the surface of CD4(+)CD8(+) T cells and natural killer (NK) cells, CD223 shares structural similarities with CD4, featuring four extracellular immunoglobulin-like domains that facilitate interaction with MHC class II. This structural homology is significant as CD223 modulates T cell responses, particularly by downregulating T cell receptor signaling, which is vital for maintaining immune homeostasis and preventing overactive immune responses. The cytoplasmic domain of CD223 is instrumental in inhibiting CD4-dependent T cell function, with the conserved Lys-468 within the "KIEELE" motif being essential for interaction with downstream signaling molecules. Understanding CD223 function and interactions is important for developing therapeutic strategies aimed at modulating immune responses in various diseases, including cancer and autoimmune disorders.
