
CD22 Antibody (RFB-4) is a mouse monoclonal IgG1 antibody that detects CD22 in human samples through immunofluorescence (IF) and flow cytometry (FCM) applications. CD22 is a critical B lymphocyte-specific antigen, also known as B-lymphocyte cell adhesion molecule (BLCAM), sialic acid-binding Ig-like lectin 2 (Siglec-2), and Leu-14. This type I integral membrane glycoprotein plays a vital role in regulating B cell signaling, which is essential for proper immune responses. CD22 is expressed in both cytoplasmic and membrane forms during various stages of B cell differentiation; the cytoplasmic variant is particularly significant as a marker for acute lymphocytic leukemia, while the membrane form is present in mature B cells before differentiation into plasma cells. Alternative splicing of the CD22 gene results in two isoforms, CD22α and CD22β, with CD22β being the predominant form. CD22 can exist as a heterodimer composed of CD22β and the shorter isoform, CD22α, highlighting the complexity of its structure and potential for diverse functional interactions within the immune system.
