CD137L Antibody (G-10) is a mouse monoclonal IgM antibody that detects CD137L protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-CD137L antibody (G-10) is available as the non-conjugated form. CD137L protein, also known as 4-1BB ligand, plays a crucial role in the immune system by providing a costimulatory signal necessary for T cell activation and proliferation. CD137L is primarily expressed on antigen-presenting cells, such as B cells, macrophages, and dendritic cells, and interacts with CD137, which is found on activated T cells. This interaction enhances immune response, particularly in tumor immunity and chronic infections. Additionally, CD137L binding to CD137 can lead to apoptosis induction in resting lymphocytes, while promoting peripheral monocyte survival through specific cytokine release. Soluble forms of CD137L have been detected in sera of patients with autoimmune conditions like rheumatoid arthritis, suggesting a potential regulatory role in modulating immune responses. CD137L′s influence on both T cell activation and monocyte survival underscores its importance in maintaining immune homeostasis and potential as a therapeutic target in immunological diseases.

Why this antibody detects a 60 KDa protein? I have reviewed other antibodies and they report a molecular weight of approximately 20 KDa. How is possible this difference?

Thank you very much for your question. CD137L is a heavily glycosylated transmembrane protein consisting of 309 amino acids. The molecular weight expected to be observed in a standard Western Blot experiment is roughly around 60 kDa as shown in the WB images on the product sheet and website. For sc-65279 we show a much smaller WB band because this is a truncated recombinant protein fragment with a much smaller size.
