CBLL1 Antibody (3B12) is a mouse monoclonal IgG2a kappa light chain antibody that detects CBLL1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-CBLL1 antibody (3B12) is available as a non-conjugated format. CBLL1, also known as Casitas B-lineage lymphoma-transforming sequence-like protein 1, HAKAI, or RING finger protein 188, is a 491 amino acid protein characterized by the presence of one C2H2-type zinc finger and one RING-type zinc finger. CBLL1 functions as an E3 ubiquitin-protein ligase, facilitating the transfer of ubiquitin from an E2 ubiquitin-conjugating enzyme to target proteins for degradation. Upon activation of c-Src, CBLL1 interacts with and ubiquitinates tyrosine-phosphorylated E-cadherin, leading to the endocytosis of the E-cadherin complex and the disruption of epithelial cell-cell contacts. This regulatory function of CBLL1 on E-cadherin is vital for maintaining cell adhesion and may also influence the processes of epithelial-mesenchymal transition, which are critical in development and cancer progression.

I am using CBLL1 (3B12): sc-517157 for immunofluorescence, what dilution of this antibody should I use?

Asked by: Randy McDonald
Thank you for your question. We recommend a starting dilution of 1:50. However, optimal antibody concentration should be determined by titration. Please find our full immunofluorescence protocol here: https://www.scbt.com/scbt/resources/protocols/immunofluorescence-cell-staining

Rated 1 out of 5 by from Poor antibodyWe bought this a month ago. Western blotting with this antibody produced a ladder of bands. Have tried multiple antibody dilutions and different antibody dilution buffers (NFDM, BSA) in more than 2 human cell lines with little improvement in results.
