cathepsin L Antibody (G-11) is a mouse monoclonal IgG2a kappa light chain antibody designed to recognize an epitope mapping between amino acids 263-291 at the C-terminus of cathepsin L, also known as CTSL, major excreted protein (MEP), or CATL. anti-cathepsin L antibody (G-11) demonstrates strong reactivity with cathepsin L from both mouse and rat species, making it ideal for applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Cathepsin L is a lysosomal cysteine protease that plays a crucial role in the intracellular degradation of proteins including collagen, elastin, and α-1 protease inhibitor, which is essential for maintaining cellular metabolism and homeostasis. Structurally, cathepsin L functions as a dimer composed of disulfide-linked heavy and light chains derived from a single precursor protein, a configuration that is vital for enzymatic activity and stability. Post-translational modifications of cathepsin L, such as glycosylation and proteolytic processing from procathepsin L to its active form, are critical for proper localization within the lysosome and regulation under acidic conditions. Overnight incubation @ 4°C Samples: SH-SY5Y cell lysates 20µg/lane, 12% Gel Transferred to PVDF membrane; blocked with 5% skimmed milk for 1hr @ RT. was supposed to detect both full length & cleaved Cathepsin L.. But doesn’t detect the low mol wt cleaved fragments. Detects protein at ~36kDa (pro cathepsin)
