Cas-L Antibody (2G9) is a mouse monoclonal IgG1 kappa light chain antibody that detects Cas-L protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Cas-L (2G9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Cas-L protein, also known as human enhancer of filamentation (HEF1) and neural precursor cell expressed, developmentally down-regulated 9 (NEDD9), plays a crucial role in integrin and growth factor signaling pathways that regulate essential cellular processes such as growth, motility, and apoptosis. Notably, Cas-L is involved in phosphorylation processes, which are influenced by cell adhesion and Src kinase activity, leading to two isoforms, p105 and p115, with the larger isoform resulting from Ser/Thr phosphorylation. This post-translational modification affects Cas-L function and interactions, particularly in the invasive behavior of glioblastoma cells, where Cas-L acts as a downstream effector of focal adhesion kinase (FAK). Furthermore, TGFβ1 regulation of Cas-L gene expression and adhesion-dependent organization of actin highlight Cas-L importance in cellular dynamics and proteasomal turnover, mediated by protein phosphatase 2A (PP2A). Tyrosine phosphorylated Cas-L binds FAK in dendrites and soma of neurons following ischemic events, and promotes neurite outgrowth in PC-12 cells, underscoring Cas-L potential role in neurodevelopment and repair mechanisms.
