CAS Antibody (H-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects CAS protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). CAS (H-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The Cellular Apoptosis Susceptibility protein (CAS), also known as Exportin 2, is a crucial 971 amino acid member of the CSE1 family that plays a vital role in the nuclear transport of proteins. Specifically, CAS mediates the re-export of Importin α from the nucleus to the cytoplasm after releasing its cargo into the nucleoplasm. This process is essential for maintaining cellular homeostasis and regulating apoptosis, as CAS ensures proteins are properly shuttled in and out of the nucleus, thereby influencing gene expression and cell survival. In the nucleus, CAS forms a complex with Importin α and the GTPase Ran in its active GTP-bound form, facilitating the docking of this complex to the nuclear pore complex through interactions with nucleoporins. Once in the cytoplasm, the complex dissociates, allowing Importin α to be released while CAS returns to the nucleus to repeat the cycle. Notably, CAS shows high expression in proliferating cells, and three isoforms of CAS have been identified due to alternative splicing, with Isoform 1 being the full-length protein, while Isoforms 2 and 3 exhibit variations in their amino acid sequences and lengths.

it is necessary a fixation method for flow citometry with CAS9 (7a9-3a3) sc 517386 right?

Thank you for your question. You can find our recommended protocol under this link: https://www.scbt.com/resources/protocols/flow-cytometry . Fixation may not be required, but can produce better results compared to live cell staining.

What is the recommended fixation method for immunofluorescence staining with CAS (H-2): sc-271537 monoclonal antibody?
