Calpain reg Antibody (P-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects Calpain reg protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Anti-Calpain reg antibody (P-1) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Calpain reg protein plays a crucial role as a regulatory subunit in the calpain family of calcium-dependent cysteine proteases, which are essential for mediating specific calcium-dependent cellular processes such as cell fusion, mitosis, and meiosis. These proteases are heterodimers composed of a small regulatory subunit, Calpain reg, and one of three large catalytic subunits: Calpain 1, Calpain 2, or Calpain p94. The regulation of calpain activity is influenced by calpastatin, which inhibits both proteolytic activity of calpain and membrane binding. This regulatory mechanism is particularly important in various physiological processes, including spermatogenesis and the acrosome reaction, which precedes fertilization. Additionally, Calpain 1 co-localizes with human leukocyte antigen-DR (HLA-DR) on activated microglia in the aging brain, highlighting potential involvement in neuroinflammatory responses. Calpain reg (P-1) antibody effectively detects and characterizes Calpain reg protein, making this antibody an invaluable tool for researchers studying the intricate roles of calpains in cellular signaling and pathology.

If I use this antibody coupled to agarose beads for IP, does the light chains appears on the western blot  where I want to reveal the immunoprecipitated CAPNS1 (same size as the light chain) ? Thanks for your response

Thank you for your question. The agarose conjugated antibody (sc-32325 AC) is covalently bound to sepharose (agarose) beads. This will greatly reduce the amount of IP antibody (heavy and light chains) which might be detected in the sample by Western blot. However, cross-linking of the IgG to the agarose will never be 100% and some antibodies are not cross-linked and may come off under elution or when using reducing agents in the sample buffer before gel loading. One way to avoid detecting the light chain IgG in your Western blot is to use the primary antibody directly conjugated to HRP (sc-32325 HRP) for direct Western blot of your immunoprecipitated samples. Then it will not be necessary to use anti-mouse IgG secondary antibody for detection.

Are there any issues with using sc-32325: Calpain reg (P-1) mouse monoclonal primary antibody on mouse samples?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent,we recommend using a directly conjugated primary antibody.
