    Calpain 7 Antibody (D-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects Calpain 7 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Calpain 7 (D-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Calpain 7, also known as PalBH, is a member of the Non-EF-hand subfamily of calpains and is characterized by a unique structure, including a long N-terminal domain and a PalB homologous domain flanking the calpain protease domain. This atypical calpain, consisting of 813 amino acid residues, exhibits only 26-35% shared identity with other calpain family members, primarily within the protease domain. The distinct structure lacks domain IV, preventing dimer formation with the 30 kDa regulatory subunit, which is crucial for typical calpain activation. Calpain 7 is predominantly expressed in the brain and localizes in both cytoplasm and nucleus, with activated forms found exclusively in the nucleus. Upregulation of Calpain 7 in striatal or cortical tissue of Huntington′s Disease knock-in mice suggests a potential role in disease onset, highlighting the importance of understanding cellular pathway functions and interactions.

    We recommend loading 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane.
