Calpain 1 Antibody (D-11) is a mouse monoclonal IgG1 kappa light chain antibody that detects Calpain 1 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Anti-Calpain 1 antibody (D-11) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Calpain 1, also known as µ-calpain, is an intracellular calcium-dependent protease that plays a crucial role in cleaving cytoskeletal and submembranous proteins, which is essential for maintaining cellular structure and function. Calpain 1 is primarily located in the cytoplasm and is activated by calcium ions, allowing participation in various cellular processes such as cell cycle regulation, apoptosis, and signal transduction. Proper functioning of Calpain 1 is vital for processes like cell fusion and spermatogenesis, as Calpain 1 influences the acrosome reaction, a key event in fertilization. Dysregulation of Calpain 1 activity has been implicated in several pathological conditions, including neurodegenerative diseases and cancer, making Calpain 1 a significant target for therapeutic intervention. Thank you for your question. This monoclonal antibody is specific for an epitope mapping between amino acids 8-33 near the N-terminus of Calpain 1 of human origin. The exact epitope has not been determined.

Gracias por tu pregunta. Según fuentes en línea, Calpaina 1 existe como una forma activa de 78 kDa unida a la membrana y una forma activa intracelular de 75 kDa. Calpaína 1 (D-11): sc-271313 se generó contra un péptido corto que abarca los dos sitios de escisión. Sin embargo, dado que no caracterizamos el sitio de unión exacto de este anticuerpo, no se sabe a qué forma activa se une. Nuestro análisis de WB muestra una banda única, pero como las dos formas activas son similares en peso molecular, es difícil determinar qué forma está detectando el anticuerpo. According to online sources, Calpain 1 exists as a membrane-bound 78 kDa active form and an intracellular 75 kDa active form. Calpain 1 (D-11): sc-271313 was raised against a short peptide spanning the two cleavage sites. However, since we do not characterize the exact binding site of this antibody, it is uncertain which active form it binds to. Our WB analysis shows a single band but since the two active forms are similar in molecular weight, it is hard to determine which form the antibody is detecting. Further immunostaining studies may need to be performed.
