Brm Antibody (E-6) is a mouse monoclonal IgG2a kappa light chain antibody that detects Brm protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Brm (E-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The Brahma protein (Brm) serves as an ATPase subunit of the Drosophila melanogaster Brm complex, which is closely related to the mammalian SWI/SNF chromatin-remodeling complex. Brm plays a crucial role as a transcriptional activator of Hox genes, interacting with nearly all transcriptionally active chromatin in a manner that does not overlap with Polycomb, a known repressor of Hox gene transcription. This unique interaction is vital for the regulation of gene expression during development, as the Brm complex acts as an essential coactivator for the trithorax group protein Zeste, which binds to DNA and activates homeotic genes. Notably, a reduction in Brm function significantly decreases the association of RNA polymerase II with Drosophila salivary gland chromosomes, indicating that chromatin remodeling activity of the Brm complex is fundamental in facilitating transcription by RNA polymerase II. Furthermore, Brm acts as a dominant suppressor of the rough eye phenotype caused by a hypomorphic mutation of Drosophila cyclin E, inhibiting S phase entry by functioning downstream of cyclin E protein accumulation. The interaction of the Brm complex with chromatin may also be influenced by BAP111, which is closely associated with the Brm complex in Drosophila embryos through an HMG domain. Brm is highly expressed in unfertilized eggs and early embryos, underscoring its importance in early developmental processes.

When should the TransCruz reagent sc-166579 X be used instead of the standard form sc-166579?

Thank you for your question. We recommend using the TransCruz reagent form when performing ChIP or GelShift assays. This form can also be used in other applications such as Western Blot or IHC, but the dilution should be adjusted by a factor of 10 to account for the higher original concentration.
