BICD2 (bicaudal D homolog 2) encodes a dynein–dynactin adaptor that couples cargo to microtubule-based minus-end transport, supporting long-range trafficking in polarized cells such as neurons. By linking vesicles, endosomes, and organelles to cytoplasmic dynein, BICD2 contributes to Golgi organization, endosome positioning, nuclear envelope dynamics, and cell cycle-related processes including spindle orientation. BICD2-dependent transport intersects with pathways governing axonal homeostasis and membrane traffic, where precise cargo delivery is required for neuronal maintenance. Genetic variation in BICD2 has been associated with neuromuscular phenotypes, making it a relevant target for studying motor neuron biology and intracellular transport defects.
BICD2 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the BICD2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the BICD2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the BICD2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish BICD2 protein expression.
