vorrei sapere cosa sta a significare la doppia banda che appare in western blot, quando si va a vedere la beta catenina. La banda di sopra e quella di sotto rappresentano la forma pro- e attiva rispettivamente della proteina o magari una è la fosforilata

Thank you very much for your inquiry. The human β-catenin full-length protein consists of 781 amino acids with an expected molecular weight around 92kDa to be observed in a Western Blot. The protein migration in the gel will be affected by different post-translational modifications, including phosphorylation, so that the actual band size observed may differ from the predicted size. Also a doublet or multiple bands might be observed occasionally depending on the Sample material and conditions used to run the actual gel. The shown doublet band might represent a phosphorylation but this was not further analyzed. In order to do this one could use a combined IP/WB approach with phospho-specific antibodies against β-catenin p-lated Tyr 654 or Tyr 86 or Ser 33/37 and of course against p-lated amino acids such as e.g. P-Tyr PY99 sc-7020.
