β-Arrestin-1/2 Antibody (A-1) is a mouse monoclonal IgG2a kappa light chain antibody designed to detect both β-Arrestin-1 and β-Arrestin-2 across multiple species including mouse, rat, human, bovine, and porcine. β-Arrestin-1/2 monoclonal antibody (A-1) is raised against amino acids 7-290 near the N-terminus of the human β-Arrestin-1 protein, ensuring reliable detection in applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). β-Arrestin-1 and β-Arrestin-2 are pivotal regulators of G protein-coupled receptor (GPCR) signaling, mediating receptor desensitization, internalization, and initiating alternative signaling pathways independent of G proteins. Structurally, both proteins consist of two distinct domains, the N-terminal domain and the C-terminal domain, which form a bilobal structure essential for their interaction with phosphorylated GPCRs and various signaling proteins. Post-translational modifications, such as phosphorylation and ubiquitination, play a significant role in modulating the functionality of β-Arrestins by regulating their interactions, stability, and localization within the cell. β-Arrestin-1 and β-Arrestin-2 interact with specific proteins including G protein-coupled receptor kinases (GRKs), clathrin, and the AP-2 adaptor complex, which are vital for receptor internalization and the formation of signaling complexes. These interactions are crucial for maintaining precise control over cellular responses to hormones, neurotransmitters, and sensory stimuli, thereby influencing processes such as hormone signaling, neurotransmission, and immune responses. Predominantly expressed in spleen and neuronal tissues, β-Arrestin-1 and β-Arrestin-2 are integral to maintaining cellular homeostasis and facilitating appropriate responses to extracellular stimuli. Anti-β-Arrestin-1/2 antibody (A-1) is recommended for researchers studying the structural and functional dynamics of β-Arrestin-mediated signaling pathways.
