Bax Antibody (B-9) is a mouse monoclonal IgG2b kappa light chain antibody that detects Bax protein of mouse, rat, and human origin through applications such as western blotting, immunoprecipitation, immunofluorescence, immunohistochemistry, and flow cytometry. Bax (B-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, phycoerythrin, fluorescein isothiocyanate, and multiple Alexa Fluor® conjugates. Bax protein plays a crucial role in the regulation of apoptosis, acting as a pro-apoptotic member of the Bcl-2 protein family. By promoting the release of cytochrome c from the mitochondria, Bax facilitates the activation of caspases, which are essential for the execution of programmed cell death. This function is particularly important in maintaining cellular homeostasis and preventing uncontrolled cell proliferation, which can lead to cancer. Notably, Bax can form both homodimers and heterodimers with Bcl-2, a well-known anti-apoptotic protein, thereby influencing the balance between cell survival and death. Bax (B-9) antibody specificity ensures detection of Bax without cross-reactivity with Bcl-2, making this reagent a valuable tool for studying apoptotic pathways. Overexpression of Bax has been shown to accelerate apoptotic death in response to cytokine deprivation, highlighting its significance in cellular responses to stress and growth factor withdrawal.

 I run my sample on 10 % gel, I used 1/1000 in 5% BSA in TBS with  3h incubation, and I didn’t get any bands. Would you please advice me. 1)  What is the recommended dilution? The  dilute should be milk or BSA?  2) How long should I Incubate?What gel %?

Thank you for your question. We would recommend using the starting dilution for Western blot (1:200) and blocking your membrane with, and incubating the antibodies in 5% nonfat milk in TBST. I have three questions: 1) What gel percentage do you use to separate Baxα and Baxβ? 2) Does Bax Antibody (B-9): sc-7480 works for IF like the (6A7) clone: sc-23959? 3) If so, in human, in mouse cells or both species' cell lines?

Thank you for your question. We are running a 15% SDS-PAGE gel for detection of Bax with this antibody. Both antibodies are recommended for IF on mouse, rat and human samples.

This is shown by knock out studies and comparison with other anti-Bax antibodies. Please see a recent article by Entrop, K. et al., 2024, Cell Death & Disease: https://www.nature.com/articles/s41419-024-07273-6 I urge SCBT to stop offering this antibody until it has been verified by knock out studies that the antibody is specific.
