Band 3 Antibody (A-6) is a mouse monoclonal IgG2a kappa light chain antibody that detects band 3 protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Band 3 (A-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Band 3, also known as AE1, is a crucial erythrocyte membrane glycoprotein that plays a vital role in maintaining cell structural integrity and facilitating the exchange of chloride and bicarbonate ions across the phospholipid bilayer. Band 3 (A-6) monoclonal antibody helps study protein function essential for proper gas exchange and pH regulation in blood, as band 3 helps transport carbon dioxide from tissues to lungs while maintaining acid-base balance. This approximately 900 amino acid protein features two distinct domains: the amino-terminal domain, or cytoplasmic domain of erythrocyte membrane band 3 (cdb3), which serves as an attachment site for erythrocyte cytoskeleton by binding to ankyrin, and the carboxy-terminal, membrane-associated domain responsible for anion exchange transport. Degradation of band 3 can lead to senescent cell antigen (SCA) formation, expressed on aging cells and signals their removal by the immune system, highlighting band 3′s role in cellular aging and immune response. An isoform of band 3 lacking the first 65 amino acids and not binding ankyrin is expressed in the kidney, indicating diverse functional implications across different tissues.
