Bag-5 Antibody (F-9) is a mouse monoclonal IgG2a kappa light chain antibody that detects Bag-5 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Bag-5 (F-9) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Bag-5, also known as Bcl-2-associated athanogene 5, is a crucial member of the Bag family of proteins, characterized by the presence of four Bag domains. These domains enable Bag-5 to bind with high affinity to the ATPase domain of HSC 70/HSP 70, thereby regulating chaperone activity and apoptosis. Bag-5 plays a significant role in the HSP 70/Parkin complex, where Bag-5 inhibits the E3 ubiquitin ligase activity of Parkin and modulates HSP 70 chaperone activity. This interaction is vital for cellular stress responses, particularly in dopaminergic neurons, as Bag-5 expression increases following neuronal injury, sensitizing these cells to injury-induced cell death. Understanding Bag-5 function is essential, as Bag-5 may serve as a therapeutic target in neurodegenerative diseases, including Parkinson′s disease, which is linked to mutations in the Parkin gene.
