ASTL Antibody (D-8) is a mouse monoclonal IgG2a kappa light chain antibody that detects ASTL protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). ASTL (D-8) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. ASTL protein, also known as astacin-like metalloendopeptidase or ovastacin, is a 435 amino acid protein that plays a crucial role in fertilization by regulating the zona pellucida, a glycoprotein layer surrounding the oocyte. This regulation prevents polyspermy, ensuring that only one sperm fertilizes the egg, which is essential for proper embryonic development. Highly expressed in unfertilized oocytes, ASTL levels drop significantly upon fertilization, indicating specific function during early stages of reproduction. Human ASTL protein shares 78% sequence identity with mouse counterpart and features an N-terminal signal peptide, a prodomain, a zinc-dependent metalloprotease domain, and a C-terminal extension that is likely heavily O-glycosylated. ASTL gene is located on chromosome 2q11.2, and two isoforms of ASTL2 arise from alternative splicing events. ASTL′s catalytic activity is inhibited by EDTA and the broad-spectrum metalloproteinase inhibitor batimastat (BB-94), highlighting potential as a target for therapeutic interventions in reproductive health.

We recommend using one of our our exclusive Mouse IgG Binding Proteins conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems.
