ASGPR1 Antibody (8D7) is a mouse monoclonal IgG1 kappa light chain antibody that detects ASGPR1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and flow cytometry (FCM). ASGPR1 (8D7) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The Asialoglycoprotein Receptor (ASGPR), also known as hepatic lectin, is a type II integral membrane protein predominantly expressed in hepatic cells, where ASGPR plays a crucial role in glycoprotein clearance from circulation. ASGPR1 is composed of two homologous subunits, ASGPR1 and ASGPR2, which form multimeric complexes and contain four functional domains: a cytosolic domain, a transmembrane domain, a stalk domain, and a carbohydrate recognition domain (CRD). The CRD enables ASGPR to specifically bind glycoproteins with terminal galactose and N-acetylgalactosamine residues in calcium′s presence, facilitating ASGPR-glycoprotein complex internalization into cells. Upon internalization, receptor and ligand dissociate, allowing ASGPR to recycle back to cell membrane. ASGPR can interact with hepatitis B virus (HBV), mediating HBV entry into liver cells, highlighting ASGPR′s potential as a therapeutic target in HBV infection.

Rated 4 out of 5 by from Test on bovine samplesThe product has been tested in western blot against samples derived from bovine plasma and bovine liver extract. Tests were run at 1:1000 and 1:500 dilutions in PBS + 0.05% tween-20 + 1% fat-free milk. As stated in the datasheet, the antibody should detect a band around 46 kDa in mouse, rat and human samples. The reactivity on bovine samples has never been tested. In our experiments, we detected a band between 37 kDa and 25 kDa. It is worth to notice that the same antibody purchased by another company gave us similar results. Moreover, the molecular weight of the protein has been predicted to be around 33 kDa in human, rat, mouse and bovine. We could detect a strong clear signal in bovine liver extract; a fainter but clear band was also present in samples derived from bovine plasma. The antibody has also been tested for immunoprecipitation of bovine plasma samples. In this case, ASGR1 detection in western blot was performed using a polyclonal rabbit antibody. The same band between 37 kDa and 25 kDa was detected.
