ASC Antibody (F-9) is a mouse monoclonal IgG1 kappa light chain antibody that targets the apoptosis-associated speck-like protein containing a CARD (ASC), also known as TMS1 or PYCARD. Anti-ASC antibody (F-9) is raised against the N-terminal amino acids 1-120 of human ASC and is effective in detecting ASC proteins from mouse, rat, and human sources using techniques such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). ASC is a critical member of the CARD-containing adaptor protein family, playing a pivotal role in apoptotic signaling by mediating interactions between adaptor proteins like Apaf-1 and pro-caspases such as CASP9. Structurally, ASC comprises a Pyrin-like domain (PYD) at the N-terminus and an 87-residue caspase recruitment domain (CARD) at the C-terminus, which are essential for its function in apoptosis regulation. Upon induction of apoptosis, ASC undergoes a transformation from diffuse cytoplasmic distribution to forming punctate or spherical perinuclear aggregates, a process that can be visualized through fluorescence microscopy. This aggregation is significant in the cellular response to apoptotic stimuli, including treatments with retinoic acid and various anti-tumor drugs. Additionally, ASC expression has been observed in multiple leukemia and melanoma cell lines, indicating potential as a biomarker in cancer research. Available in both non-conjugated and various conjugated forms, including horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® variants, ASC (F-9) antibody serves as a versatile and essential tool for researchers studying apoptotic mechanisms and related cellular pathways.

We can see a number of ASC specks in human macrophages treated with ATP control, but not untreated cells when cells were stained with this antibody after treatment. I recommend it!
