Arginase 1 Antibody (E-2) is a mouse monoclonal IgG1 kappa light chain antibody engineered to detect Arginase 1 of mouse and rat origin across multiple applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-Arginase 1 antibody (E-2) is designed to recognize a precise epitope mapped between amino acids 43-81 near the N-terminus of mouse Arginase 1, ensuring accurate and reliable binding. Arginase 1, also called ARG1, AI, Arg-1, and PGIF, is predominantly expressed in the liver, and plays a critical role in the urea cycle by catalyzing the conversion of arginine into ornithine and urea, thereby facilitating the detoxification of ammonia and maintaining nitrogen balance within the body. Structurally, Arginase 1 functions as a homotrimeric protein, each monomer containing a binuclear manganese cluster that is essential for enzymatic activity. This trimeric structure not only provides the necessary catalytic sites for efficient substrate conversion but also contributes to enzyme stability and regulatory mechanisms. Understanding the structural intricacies of Arginase 1 is vital as this knowledge underpins functionality in metabolic pathways and involvement in various physiological processes such as wound healing, immune response modulation, and cancer progression. Can this antibody be used for human cell line western blotting experiment?

Thank you for your question. Anti-liver Arginase 1/ARG1 Antibody (E-2): sc-271430 is recommended for detection of Arginase 1 of mouse and rat origin by WB, IP, IF, IHC(P) and ELISA, not recommended for human cell line western blotting experiment.

I performed the western blotting using arginase I antibody (E-2). But M1 macrophage showed two band between 35~50 kDa and M2 macrophage showed one band between 35~50 kDa. Question: In case of two band, all two band are arginase 1? or isoform can be?

Does this antibody specifically recognize Arg-1, or is there some cross reactivity with Arginase-2?

Thank you for your question. This mouse monoclonal antibody is raised against an antigen that is specific for mouse and rat arginase I, and does not cross react with human. The antigen is specific for arginase I, with no significant similarity to arginase II.

If the level of expression is very high, the Ab works perfectly. If the Arginase1 level is low, signal could be missed as there is some background present.

Rated 2 out of 5 by from Western blot analysisNOS II and Arg-1 were detected using the appropriate antibodies at a dilution of 1:750 and 1:1000, respectively (Santa Cruz Biotechnology, CA, USA), with overnight incubation at 4 °C. Goat anti-rabbit peroxidase-conjugated antibody and goat anti-mouse peroxidase-conjugated antibody (Jackson ImmunoResearch; West Grove, PA, USA) were used at a 1:25,000 dilution as the secondary antibody. Membranes were stripped and incubated with anti-β-tubulin antibody (Santa Cruz Biotechnology CA, USA) at a dilution of 1:2,500 as a loading control.
