ARC Antibody (A-2) is a mouse monoclonal IgG2a antibody that detects ARC in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). ARC, also known as apoptosis repressor with CARD domain or nucleolar protein 3 (NOL3, NOP, NOP30), plays a crucial role in regulating apoptosis by inhibiting caspases, specifically caspase-2 and caspase-8, which are key mediators of programmed cell death. Anti-ARC antibody (A-2) recognizes a protein that prevents apoptosis, which is vital for cellular survival, particularly in tissues such as skeletal muscle and heart, where ARC is expressed in two transcript variants of 5.5 kb and 1.0 kb. Notably, the 1.0 kb transcript has been shown to inhibit apoptosis in a dose-dependent manner when coexpressed with caspase-8, highlighting its protective function in cellular contexts where apoptosis is triggered by external signals like CD95/FAS and tumor necrosis factor receptor-1. Furthermore, ARC is phosphorylated at Threonine 149 by casein kinase 2 (CK2), a modification that is essential for its localization to the mitochondria, thereby enhancing its role in apoptosis regulation. ARC monoclonal antibody (A-2) serves as an invaluable tool for researchers studying apoptosis and its implications in various diseases.
