AQP2 encodes aquaporin-2, a vasopressin-regulated water channel that controls osmotic water reabsorption in renal collecting duct principal cells. In response to AVPR2–cAMP–PKA signaling, AQP2 undergoes phosphorylation-dependent trafficking to the apical membrane, coordinating with endocytic recycling and cytoskeletal dynamics to tune membrane water permeability. This pathway integrates hormonal control of fluid balance with epithelial polarity and membrane transport processes. Dysregulation of AQP2 expression, localization, or gating is linked to impaired urinary concentrating ability and serves as a mechanistic node in studies of water homeostasis and renal tubular dysfunction.
Aquaporin 2/AQP2 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the AQP2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the AQP2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the AQP2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Aquaporin 2/AQP2 protein expression.
