ANP32A/B Antibody (A-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects ANP32A/B protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). ANP32A/B (A-2) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. ANP32A protein, also known as Inhibitor 1 of protein phosphatase 2A (I1PP2A), plays a critical role in regulating protein phosphatase 2A (PP2A) activity, which is essential for numerous cellular processes, including cell growth, division, and apoptosis. By inhibiting PP2A, ANP32A modulates signaling pathways that are vital for maintaining cellular homeostasis and preventing uncontrolled cell proliferation. ANP32B, a 251 amino acid protein also referred to as acidic (leucine-rich) nuclear phosphoprotein 32 family member B, is predominantly localized in the nucleus and contains four leucine-rich repeat motifs. ANP32B is expressed in various tissues, including the placenta, heart, pancreas, lung, prostate, spleen, and thymus, and is crucial for cell cycle progression, particularly in facilitating the transition from the G1 to S phase. ANP32B acts as an anti-apoptotic factor by inhibiting caspase-3 activity, thereby playing a significant role in cell survival. Anti-ANP32A/B antibody (A-2) versatility in various applications makes this reagent an invaluable tool for researchers studying the intricate roles of these proteins in cellular regulation and disease mechanisms.

Can ANP32A/B (A-2): sc-374552 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: DefinitelyNotMatt
Thank you for your question. Yes, ANP32A/B (A-2): sc-374552 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 5 out of 5 by from Highly potent and specific antibodyThis monoclonal antibody was independently tested in-house by Kinexus Bioinformatics Corporation and validated for use for Western blotting. There was very strong detection of the ~29 kDa target protein in HeLa/Jurkat cell lysate mix, but target not detected in mouse tissues (brain/testes/thymus/muscle mix). No major cross-reactivities were observed at 2 µg/ml antibody concentration with off-target proteins in the mouse tissues tested or in the HeLa/Jurkat cell lysate mix.
