Anillin Antibody (B-10) is a mouse monoclonal IgG2b kappa light chain antibody that detects Anillin protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-Anillin antibody (B-10) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Anillin, also known as scraps homolog, is an evolutionarily conserved actin-binding protein that plays a crucial role in cytokinesis, a process essential for cell division. Anillin is predominantly localized in the nucleus and is highly expressed in the central nervous system, where Anillin localizes to the cleavage furrow during cytokinesis, forming a contractile ring with the assistance of Rac GTPase. The interaction of Anillin with CD2AP is vital for concentrating Rho A and ensuring proper localization of active Myosin, which is necessary for successful cleavage furrow ingression. Notably, in Anillin knockout cells, the cleavage furrow fails to complete ingression, highlighting Anillin′s critical function in cell division. Anillin expression levels are tightly regulated throughout the cell cycle, peaking during mitosis, and before cell transition into G1, Anillin associates with E-cadherin and undergoes ubiquitination by the anaphase-promoting complex/cyclosome (APC/C), which recognizes the D-box domain at Anillin′s N-terminal region. Given Anillin′s overexpression in various tumors, Anillin may serve as a potential biomarker for cancer diagnosis and prognosis, making Anillin (B-10) antibody an invaluable tool for researchers studying cell division and cancer biology implications.

Can Anillin (B-10): sc-271814 monoclonal antibody be used to stain paraformaldehyde-fixed cancer cells for immunofluoresence microscopy? Thank you!

Thank you for the question. We have not yet tested this antibody on pfa- fixed cells for IF. Instead we suggest using cells fixed with cold methanol according to our protocols at a starting dilution of 1:50. Since this antibody is working well in IHC(P) on formalin-fixed, paraffin-embedded tissue sections, it is not unlikely this clone can also be used to stain paraformaldehyde-fixed cancer cells for immunofluoresence microscopy. Can Anillin (B-10): sc-271814 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Thank you for your question. Yes, Anillin (B-10): sc-271814 has been validated for use in IHC with paraffin-embedded sections. Rated 5 out of 5 by from Efficient WB antibodyI got a free aliquot of this antibody and tested it on HEK293 samples transfected with siCo or siANLN. The antibody showed high specificity with no background. I used it 1:1000 in 0,1% milk with O/N incubation at 4°C. I would recommend it.
