Ameloblastin Antibody (H-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects Ameloblastin of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Anti-Ameloblastin antibody (H-2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Ameloblastin plays a crucial role in dental enamel development, a highly mineralized tissue primarily composed of hydroxyapatite crystals. Ameloblastin (H-2) antibody specifically detects this protein in the extracellular matrix, where Ameloblastin is essential for proper formation and mineralization of the enamel matrix during tooth development. Ameloblastin interacts with other key proteins such as amelogenin and enamelin, contributing to the structural integrity and organization of enamel. Defects in the AMBN gene, which encodes for Ameloblastin, have been associated with conditions like ameloblastomas, highlighting Ameloblastin′s significance in dental health. Anti-Ameloblastin antibody (H-2)′s ability to effectively detect Ameloblastin across various applications makes this reagent an invaluable tool for researchers studying enamel formation and related pathologies.

Can Ameloblastin (H-2): sc-271012 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: Randy McDonald
Thank you for your question. Yes, Ameloblastin (H-2): sc-271012 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

Rated 3 out of 5 by from Works for Westerns not for IHC in mouse tissueThis antibody worked for recombinant murine and native porcine Ambn Western blots but did not work in IHC staining of mouse sections. Previous M300 ameloblastin antibody (rabbit polyclonal) used to work very well (1:500 dilution for IHC) and N18 ameloblastin ab (goat polyclonal) worked at higher concentrations (1:100). I have tried both concentrations for H2 and it did not work.

Rated 1 out of 5 by from Doesn't work in rat and mouse ameloblastSC's previous polyclonal AMBN antibody worked great for me, however this new mouse mono Ab doesn't seem to be equivalent. It doesn't work in my immunofluorescent staining of rat and mouse ameloblasts (paraffin embeding and enzyme/citrate heat retrieval).
