α-Adaptin 1/2 Antibody (AP.6) is a mouse monoclonal IgG1 kappa light chain antibody that detects α-Adaptin 1 and α-Adaptin 2, as well as an alternatively spliced isoform of α-Adaptin 1 found in neurons of mouse, rat, and human origin through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-α-Adaptin 1/2 antibody (AP.6) plays a crucial role in the assembly of clathrin-coated pits and vesicles, which are essential for receptor-mediated endocytosis, a process vital for cellular communication and nutrient uptake. α-Adaptin 1/2 is primarily located in the cytoplasm and is involved in the transport of vesicles from the trans-Golgi network and the plasma membrane, facilitated by the adapter complexes AP-1 and AP-2. These complexes consist of clathrin-binding adaptin proteins, including γ-Adaptin and β-Adaptin for AP-1, and α-Adaptin 1, α-Adaptin 2, and β2-Adaptin for AP-2, along with smaller subunits known as AP50 and AP17. The structural organization of α-Adaptin, characterized by its two-domain architecture with variable C-terminal domains, is significant as this allows for the differential regulation of clathrin-mediated endocytosis in various tissues, highlighting the importance of splice variants in maintaining cellular function. Additionally, proteins such as AP180 and CALM, which are homologous to α-Adaptin, further underscore the complexity of clathrin assembly and its implications in synaptic function and cellular trafficking.

I am using α-Adaptin 1/2 (AP.6): sc-32284 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
