α-actinin-4 Antibody (G-4) is a mouse monoclonal IgG2a kappa light chain antibody that detects alpha-actinin-4 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). anti-alpha-actinin-4 antibody (G-4) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Alpha-actinin-4 plays a crucial role in the cytoskeleton by cross-linking actin filaments, which is essential for maintaining cell shape, motility, and structural integrity. Alpha-actinin-4 is predominantly found in non-muscle cells, where alpha-actinin-4 contributes to cytoskeleton organization and is involved in various cellular processes, including cell adhesion and migration. Alpha-actinin-4′s presence in the cytoplasm and at the cell membrane underscores alpha-actinin-4′s importance in cellular dynamics and signaling pathways. Notably, mutations in the gene encoding alpha-actinin-4 have been linked to focal segmental glomerulosclerosis 1 (FSGS1), a serious kidney disorder characterized by progressive renal dysfunction, highlighting alpha-actinin-4′s significance in both health and disease.

Thank you for your question. Yes, α-actinin-4 (G-4): sc-390205 has been validated for use in IHC with paraffin-embedded sections. Rated 5 out of 5 by from Clean signal in IF, good detection on WBThis antibody worked in IF and WB both for different human melanoma cell lines (MDA-MB-435S and RPMI-7951). It showed differential expression among the cell lines and the expression was in line with the mass spectrometry results.

Rated 5 out of 5 by from Works great for Westerns and IF with PFA fixationThis antibody worked perfectly in WB at 1:1000 dilution and produced a nice clean band right at 100 kDa, showing expected differences in the expression between parental cells and the clones that we made, confirming our mass spectrometry data. It also worked great for IF staining in cultured melanoma and breast cancer cell lines after PFA fixation. The fluorescence is strong and clear.

Rated 5 out of 5 by from Works greatWorks great for Westerns. Strong bands when used at 1:500 with 10 ug whole cell extract, extremely low background, and no non-specific binding. Band showed up right at 100 kDa. Produced a nice clean band with and without adding BME to Laemmli buffer.
