ALAS-E Antibody (D-4) is a mouse monoclonal IgG1 kappa light chain antibody that detects ALAS-E of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). ALAS-E (D-4) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. ALAS-E, also known as 5-aminolevulinate synthase 2, plays a crucial role in the heme biosynthetic pathway, specifically catalyzing the first step in heme synthesis, which is vital for oxygen transport and cellular respiration. This erythroid-specific isoenzyme is essential for hematopoietic cell differentiation and iron metabolism, particularly in the liver, where ALAS-E regulates heme availability for hemoglobin production. Notably, ALAS-E interacts with succinyl CoA, which induces a conformational change facilitating mitochondrial translocation, a critical step for function. ALAS-E regulation is influenced by transcription factors such as GATA-1, NF-E2, and EKLF, which bind to the 5′-flanking region, highlighting gene expression control importance in erythropoiesis. Since ALAS-E is located on the X chromosome, mutations can lead to pyridoxine-refractory X-linked sideroblastic anemia, underscoring clinical significance in hematological disorders.
