SEE ALSO...

Akt1 Antibody (B-1) is a mouse monoclonal IgG1 kappa light chain antibody that detects Akt1 of mouse, rat, and human origin by WB, IP, IF, IHCP, FCM, and ELISA. Akt1 monoclonal antibody (B-1) is available in both non-conjugated and various conjugated forms, including agarose, HRP, PE, FITC, and multiple Alexa Fluor® conjugates. Akt1, also known as protein kinase B (PKB), plays a crucial role in cellular signaling pathways that regulate cell growth, survival, and metabolism. Akt1 is primarily located in the cytoplasm and is translocated to the plasma membrane upon activation by growth factors such as insulin and platelet-derived growth factor (PDGF). This translocation is essential for activation, as Akt1 interacts with phosphoinositide-dependent kinase-1 (PDK1), which phosphorylates Akt1 at Thr 308, a critical step for full activation. Furthermore, Akt1′s ability to phosphorylate various downstream targets is vital for mediating cellular responses to growth signals, making Akt1 a key player in processes such as glucose metabolism and apoptosis. The phosphorylation of Akt1 at Ser 473, while not required for Thr 308 phosphorylation, further enhances activity and is indicative of promoting cell survival and growth. The regulation of Akt1 is complex and involves various upstream signaling pathways, highlighting importance in both normal physiology and disease states, including cancer. These features make Akt1 monoclonal antibody (B-1) an essential tool for investigating Akt1-mediated signaling pathways and roles in cellular processes and disease research.

I recently used your antibody. I noticed that it gives me signal at two different molecular weight heights: one band is at 60 kDa while the other band is around 100 kDa. How is it possible? How can i explain it?

Among the different indications of this antibody: Akt1 (B-1): sc-5298, it is its ability to recognize the phosphorylated-AKT1. Since there are different phosphorylation sites, I need to be sure that it applies also for the S473-phosphorylated AKT1.

Asked by: stefania70
Thank you for your question. This antibody can detect the Akt1 protein, both phosphorylated and non-phosphorylated. It is not phospho-specific and does therefore not specifically detect a certain phosphorylation site. If you have any further questions, please contact our European Technical Service team. You can reach them by phone at Toll Free: +49 6221 4503 0, by email at: europe@scbt.com or by live chat directly on our website, www.scbt.com

Akt1 Antibody (B-1): sc-5298 1) Does it only binds Akt1, or also binds phosphorylated Akt1? 2) How to get a 10 ug sample? How much do we need to pay?

Asked by: tua64317
Thank you for your question. This monoclonal, Akt1 Antibody (B-1): sc-5298, is able to recognize Akt1 and phosphorylated Akt1. This is demonstrated in the Western blot data on our website showing evaluation of inactive and active human recombinant Akt1. Please contact our Technical Service Department to request a complimentary 10 µg sample of this mouse monoclonal antibody for testing. The sample is free of charge. Please call us directly at (800) 457-3801 option 2, email scbt@scbt.com, or contact us by live chat.

Rated 5 out of 5 by from sc-5298-sampleI used this sample in Western blot using my mouse NSCs. I found the result excellent even without any setup, just following the data sheet indication. I got a single band of the correct size.
