AKAP 82 Antibody (25) is a mouse monoclonal IgG1 kappa light chain antibody that detects AKAP 82 protein of mouse and rat origin by western blotting (WB) and immunoprecipitation (IP). Anti-AKAP 82 antibody (25) is available as the non-conjugated format. The AKAP (A-kinase anchoring proteins) family plays a crucial role in cellular signaling by anchoring protein kinase A (PKA) to specific cellular locations, thereby ensuring precise regulation of PKA activity. AKAP 82, also known as PRKA4 (protein kinase A-anchoring protein 4) or major sperm fibrous sheath protein, is predominantly expressed in spermatids during spermiogenesis, where AKAP 82 is essential for fibrous sheath assembly, a structure critical for sperm motility and function. AKAP 82 features two distinct binding sites for PKA, one specific for the regulatory subunit RIα and another that can accommodate either RIα or RIIα, which underscores versatility in signaling pathways. AKAP 82 interacts with AKAP 3, forming a significant component of the fibrous sheath structure. Absence of AKAP 82 in knockout models leads to a marked reduction or loss of associated proteins such as AKAP 3, RIIα, SP17, and GAPDS, which can result in compromised sperm function and may contribute to conditions like asthenospermia. AKAP 82′s ability to facilitate precise PKA localization is vital for maintaining signaling pathway fidelity that governs spermatogenesis and sperm motility.

Thank you for your question. We recommend using 1–10 µl (i.e., 0.2–2 µg) of antibody with 1 ml of cell lysate. However, optimal antibody concentration should be determined by titration.
