ADPN Antibody (C-8) is a mouse monoclonal IgG2b antibody that detects ADPN in human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). ADPN, a member of the Adiponutrin family, plays a crucial role in lipid metabolism due to its lipase activity, which is contingent upon the presence of an activated serine residue. ADPN is primarily located in adipose tissue, where ADPN is involved in regulating energy homeostasis and influencing insulin sensitivity. ADPN expression is significantly affected by metabolic states; for instance, D-glucose can induce a seven-fold increase in ADPN mRNA levels, while insulin has a more modest effect. Notably, the glucose-induced increase in ADPN expression can be inhibited by factors that elevate intracellular cyclic adenosine monophosphate (cAMP) levels. Furthermore, research indicates that higher levels of ADPN mRNA are associated with improved insulin sensitivity, linking ADPN to conditions such as obesity and diabetes. ADPN gene expression in humans is tightly controlled by changes in energy balance, highlighting ADPN′s importance in metabolic health. In murine adipocytes, ADPN expression is closely aligned with that of fatty acid synthase and Srebp1c, underscoring ADPN′s role in lipid synthesis and overall metabolic regulation.
